An Optimal MPS Workflow

The PowerSeq® 46GY System contains reagents to amplify autosomal and Y-STR loci as small amplicons (140–300bp) that can be used to prepare MPS libraries and generate sequencing data compatible with Illumina® TruSeq® and MiSeq® technologies. With this workflow, both sequence and allele length polymorphism in the autosomal and Y-STR loci can be identified, thereby increasing the statistical power of inclusion. MPS also eliminates the problem of overlapping alleles completely, allowing us to target more alleles in the multiplex.

The combination of these STR loci and Amelogenin makes this multiplex an effective tool for human identification using MPS, while maintaining compatibility with existing databases worldwide. A combination of familiar markers in the PowerSeq® 46GY System and data analysis software package GeneMarker®HTS from SoftGenetics, LLC, offers a smooth transition to include MPS into your workflow, ensuring that you get the data you want.

Proper bead mixing is critical to successful purification of amplified products. This video illustrates key steps in the purification protocol.

New! This application note provides an alternate protocol for the PowerSeq® 46GY System for use of the KAPA HyperPrep Library Prep Kit for library preparation and ProNex® chemistry for bead purification.

Workflow

The PowerSeq® 46GY System integrates into your laboratory workflow without the burden of analyzing supplementary, unestablished markers—making your transition to MPS a bit easier. When using the PowerSeq® Systems, you can expect to see similar performance with regards to sensitivity, inhibitor tolerance and balance that you've come to expect from our PowerPlex® STR Systems.

Balanced Profiles

Good amplicon balance within STRs is important to prevent dropouts, resulting in improved identification of minor contributors in a mixture. Improved balance among loci and the ability to identify minor contributor alleles based on length and sequence polymorphism greatly helps with mixture interpretation.

balanced profiles for powerseq 46gy mps

The PowerSeq® 46GY System generates balanced DNA libraries. Normalized read depth per locus using 1ng 2800M Control DNA (n = 3). To control for sample loading on the MiSeq® instrument, abundance per locus was normalized to the average of total abundance for the sample set. A pool of 96 samples was sequenced simultaneously using MiSeq® Reagent Kit v3, 2 × 300 cycles. Data were analyzed with GeneMarker HTS Software v 2.3 (SoftGenetics, LLC).

Reliable Sensitivity

The PowerSeq® 46GY System offers the same sensitivity that you've come to depend on when using our PowerPlex® systems. While full profiles can be obtained with as little as 62.5pg of input DNA, the balance at low template inputs (100pg) is comparable to the balance seen at the recommended input amount of 1000pg.

powerseq 46gy reliable sensitivity

The PowerSeq® 46GY System generates balanced profiles at low DNA input amounts. Percent of total allele calls per locus at different amounts of 2800M Control DNA (n = 20). A pool of 96 samples was sequenced simultaneously using MiSeq® Reagent Kit v3, 1 × 300 cycles. Data were analyzed with GeneMarker HTS Software v2.2.3 (SoftGenetics, LLC).

Large Combination of Dependable Loci

The PowerSeq® 46GY System has the largest combination of autosomal and Y-STR loci in a single kit to generate sequence data and improve your MPS workflow. Amplicons for each locus are designed to be in a range of 140–300bp. The loci included are CODIS core and European Standard Set (ESS) loci, Amelogenin and the Y-STR loci included in the PowerPlex® Y23 System.
