CellTiter-Glo® Luminescent Cell Viability Assay는 대사가 활발한 세포의 지표인 ATP를 정량하여 배양된 세포에서 살아있는 세포 수를 측정하는 homogenous assay입니다. CellTiter-Glo® Assay는 multiwell 포맷에서 사용하기에 적합하도록 디자인되어 high-throughput screening (HTS), 세포 증식 및 세포 독성 분석에 이상적입니다. 이 homogenous assay 과정은 단일 시약(CellTiter-Glo® Reagent)을 serum이 포함된 배지에 직접 처리하는 방식으로 세포 washing, 배지 제거, 반복적인 피펫팅 과정이 필요하지 않습니다. 이 제품은 384-well 포맷에서 시약을 추가하고 섞은 후 10분 이내에 well당 최소 15개의 세포를 검출할 수 있습니다.
Homogenous “add-mix-measure” 형식으로 cell lysis가 진행되고, 세포 내 ATP 양에 비례하는 발광 signal이 생성됩니다. ATP 양은 배양된 세포 수와 직접적으로 비례합니다. CellTiter-Glo® Assay는 ‘glow-type’의 발광 signal을 생성하며, 이 발광 signal의 반감기는 세포 종류와 배지에 따라 다를 수 있지만 일반적으로 5시간 이상 유지됩니다. 이러한 긴 반감기는 시약 injector 없이 여러 plate를 연속적으로 또는 배치별로 처리할 수 있는 유연성을 제공합니다. 이 고유한 homogenous 방식은 여러 단계가 필요한 다른 ATP 측정 방법에서 발생할 수 있는 오류를 방지해 줍니다.
