GoTaq® qPCR 및 RT-qPCR System은 BRYT Green® dye가 포함된 ready-to-use 2X master mix 형태로 제공됩니다. BRYT Green® Dye는 SYBR® Green I보다 더 높은 형광 강도를 가지고 있으며 PCR inhibition을 최소화하고 증폭의 효율을 극대화하는 특징을 갖고 있습니다.

GoTaq® qPCR Master Mix는 real-time quantitative PCR을 위한 ready-to-use 2X master mix입니다. GoTaq® Hot Start Polymerase, 최적화된 buffer, 그리고 BRYT Green® Dye로 구성되어 있는 GoTaq® qPCR Master Mix는 보다 빠른 정량 cycling 값과 넓은 dynamic range를 제공하며, 이를 통해 real-time quantitative PCR에서 뛰어난 신뢰성, 재현성, 그리고 민감도를 보장합니다.

qPCR master mix data — **Single-copy 검출을 이용한 qPCR** 10배 serial dilution된 human genomic DNA(0.01ng에서 100ng까지)에서 GAPDH 유전자를 증폭 및 검출하였습니다. 해당 그래프는 다양한 dilution 농도에 대한 standard curve를 보여줍니다 (Slope=-3.2; R²=0.995). 제품 수율과 순도는 흡광 방법으로 통해 측정하였습니다.

하나의 반응에서 역전사 및 qPCR 수행

GoTaq® 1-Step RT-qPCR System은 GoScript™ Reverse Transcriptase와 GoTaq® qPCR Master Mix를 하나의 reaction well에서 수행하는 1-step real-time RT-qPCR 방법으로, RNA expression level을 검출하고 상대 정량이 가능합니다. 이 제품은 1-Step RT-qPCR을 위한 GoScript™ RT Mix (50X)는 GoScript™ Reverse Transcriptase, RNasin® Plus RNase Inhibitor과 single-step 반응을 촉진하는 첨가물이 최적의 양으로 구성되어 있습니다.

1-Step System이 제공하는 기능들은 다음과 같습니다:

넓은 dynamic range에서 low-copy와 high-copy 타겟에 대한 민감한 검출, input RNA를 femtogram(fg) 수준까지 검출이 가능함.

A6020 1-Step Real-Time PCR & qPCR Kit — **100ng에서 10fg까지의 human RNA template에 대한 linear dynamic range** 10배 serial dilution한 7개의 human total RNA 샘플을 대상으로 200nM GAPDH transcript 특이적인 primer를 사용하여 RT-qPCR을 수행하였습니다. 증폭 곡선은 97%의 효율과 r²=0.999를 나타냈습니다.

2-step 역전사 및 qPCR

GoTaq® 2-Step RT-qPCR System은 GoScript™ Reverse Transcription System과 GoTaq® qPCR Master Mix를 사용하여 2-step RT-qPCR 방법을 통해 RNA expression level을 검출하고 상대 정량을 할 수 있습니다.

GoTaq® qPCR Master MixGoTaq® qPCR Master Mix는 ready-to-use, 안정화된 2X 형태로 제공되며, GoTaq® Hot Start Polymerase, MgCl₂, dNTPs, BRYT Green® Dye와 프로메가만의 독보적인 reaction buffer가 포함되어 있습니다. 이 master mix에는 낮은 농도의 carboxy-X-rhodamine(CXR)이 포함되어 있으며 별도의 튜브로 CXR reference dye가 함께 제공됩니다.

GoScript™ Reverse Transcription System은 최적화된 reaction buffer와 M-MLV reverse transcriptase가 함께 구성되어 있어 PCR 증폭을 준비하는 과정에서 효율적인 first-strand cDNA 합성을 가능합니다. 합성된 cDNA는 downstream qPCR 증폭 반응에 직접 사용할 수 있습니다.

A6010 2-Step Real-Time PCR & qPCR Kit — **확장된 linear range** Dilution한 human reference total RNA를 GoTaq® 2-Step RT-qPCR System을 사용하여 5.8S 유전자를 증폭하였습니다. GoTaq® 2-Step System은 강한 형광 신호를 통해 9log 이상의 linear dynamic range를 보여줍니다. NTC (Negative Template Control) 반응은 모두 blank 값으로 나왔습니다.
