GoScript™ Reverse Transcriptase는 Oligo(dT) Primer 또는 Random Primer가 포함된 편리한 master mix 형태, 역전사에 필요한 모든 구성품을 포함한 complete kit, 그리고 enzyme 단독 형태로 제공됩니다.

GoScript™ Reverse Transcription System은 GoScript™ Reverse Transcriptase, Reaction Buffer, MgCl₂, dNTP Mix, Oligo(dT) Primer, Random Primer 및 Recombinant RNasin® Ribonuclease Inhibitor로 구성되어 있으며 효율적인 first-strand cDNA 합성을 위해 설계되었습니다. 이 System은 total RNA, poly(A)+ mRNA 또는 합성된 transcript RNA를 포함한 다양한 RNA template을 효과적으로 역전사할 수 있습니다.

GoScript™ Reverse Transcription Mix, Oligo(dT)

GoScript™ Reverse Transcription Mix, Oligo(dT)는 first-strand cDNA 합성을 효율적으로 수행하기 위해 최적화된 reagent로 구성되어 있습니다. Oligo(dT) primer은 polyadenylated RNA 분자의 3' 말단에 결합하여 first strand 합성을 시작합니다. 이 과정에서 total RNA 또는 mRNA를 사용하여 poly(A)+RNA을 타겟으로 oligo(dT)-priming된 cDNA를 생성합니다. 이 방법을 사용하여 생성된 cDNA는 단일 역전사 반응 이후 다양한 downstream 분석을 수행할 수 있습니다.

GoScript™ Reverse Transcription Mix, Random Primers

GoScript™ Reverse Transcription Mix, Random Primers는 qPCR 증폭을 위한 first-strand cDNA 합성에 사용되는 random primer가 포함되어 있습니다. Random primer를 사용하는 priming은 다양한 RNA template에서 cDNA 합성할 때 사용하는 가장 일반적인 방법입니다. Random primer는 poly(A)+ tail을 갖지 않는 RNA와 원핵 생물에서 분리된 RNA를 포함한 모든 RNA 분자로부터 first-strand cDNA 합성하는 데 사용할 수 있습니다.

다양한 RNA input 양으로부터 효율적인 cDNA 합성

efficient cDNA Synthesis Over a Range of Input RNA 13870MB-W — GoTaq® 2-Step RT-qPCR System을 사용하여 human total RNA에서 6가지 타겟 RNA를 직접 정량했습니다. Human reference total RNA를 1/10 dilution하여 9개의 서로 다른 농도의 샘플(10ng/ul에서 0.1fg/ul까지)을 준비한 후 GoScript™ RT로 역전사를 수행하였습니다. 각각의 역전사 산물인 cDNA 10ul를 사용하여 GoTaq® qPCR 반응을 3회씩 수행했습니다. 이 qPCR 실험은 서로 다른 expression level의 RNA 타겟을 분석하기 위해 디자인하였습니다. 매우 높은 발현률(5.8S rRNA), 높은 발현률(ACTB 및 B2M), 중간 발현률(HPRT), 또는 낮은 발현률(RPL29 및 TBP1)의 각 타겟에서 RNA input 농도가 10배씩 증가할 때마다 qPCR cycle(Cq)이 약 3.3씩 변화하는 것으로 관찰되었습니다. 이 결과는 RNA 농도에 따른 정량적 변화가 정상적으로 이루어졌음을 의미합니다.

Inhibitor에 대한 저항성

GoScript reverse transcriptase resistance to inhibitors 8675MB-W — in vitro-transcribed RNA를 각 제조사의 매뉴얼에 따라 oligo(dT) primer를 사용하여 역전사를 수행하였습니다. 역전사는 에탄올 0%, 5%, 10%, 20% 조건에서 수행되었습니다. 생성된 cDNA는 GoTaq® qPCR Master Mix를 사용하여 qPCR을 수행하여 분석되었습니다. 에탄올에 따라 Cq(cycle threshold)의 값을 비교하였고, 이를 전사된 cDNA 양의 상대적 변화를 2(Cq inhibitor – Cq none)로 계산하였습니다. 데이터는 세 번 실험하여 얻은 결과의 평균값입니다.

Low-copy 타겟의 효율적인 검출

Total human RNA(Agilent, Cat #750500)의 10배 dilution한 샘플(1µg - 100pg)을 GoScript™ Reverse Transcription Mix, Random Primer 또는 Oligo(dT) Primer를 사용하여 역전사하였습니다. 생성된 cDNA는 GoTaq® qPCR Master Mix (Cat. # A6002)를 사용하여 low-copy 유전자 APC 또는 TBP를 qPCR 증폭을 통해 측정하였습니다. Standard curve에 따르면 primer의 효율은 각각 99%(APC)와 97%(TBP)로 계산되었으며 두 값 모두 MIQE 가이드라인에서 권장하는 범위 내에 있음을 보여줍니다. 역전사 효소를 포함하지 않은 negative control과 template을 포함하지 않은 control에서는 음성 결과가 나타났습니다.
